Applications of BBa_K1966001

This part has been shown to be functional for the following activities:

1)conversion of benzyl alcohol to benzaldehyde
2)conversion of 3-methylbenzyl alcohol to m-tolualdehyde
3)conversion of 4-methylbenzyl alcohol to p-tolualdehyde
4)conversion of 3-chlorobenzyl alcohol to 3-chlorotolualdehyde
5)conversion of 3-methoxybenzyl alcohol to m-anisaldehyde
-Enhancement of indigo biosynthesis

Part BB1_K1966001 is built into the composite part BBa_K1966003, which is demonstrated in the figure below. This shows functionality of BBa_K1966001, as the aromatic aldehyde is not generated without P. putida BADH activity.

The figure below shows conversion of 3-chlorobenzyl alcohol and 3-methoxybenzyl alcohol to their respective aldehydes, as well as failure to act on 4-methoxybenzyl alcohol. E. coli BL21 DE3 with part BBa_K1966001 was grown for 8 hours in the presence of each substrate.

The prescence of BADH in cultures containing P. putida xylene monooxygenase (XO, from part BBa_K1966000) has been shown by the 2016 UTK iGEM team to enhance indigo biosynthesis. This is reflected in the following figure.

Further characterization will be attempted for the following functions:

1)Conversion of 4-methoxybenzyl alcohol to p-anisaldehyde
